IT seminar: Renal cysts formation - Study on the organization and dynamics of 3D biomimetic cellular models for polycystic disease

Abstract:

We propose to develop an original kidney-on-a-chip model to mimic Autosomal Dominant Polycystic Kidney Disease (ADPKD), the most common inherited kidney disease, and to decipher the disease mechanisms with a high level of control. In ADPKD cysts form exponentially in clusters in kidney tubules after a long period of low cyst incidence and lead to kidney failure, reminding of a snowball effect. The proposed deformable microfluidic chip channels would reproduce geometrical and mechanical properties of the kidney tubules. This, with controlled cellular heterogeneity, mutations induced with optogenetics and mechanical tube compression will help understand the hypothesized snowball effect.
